Combinations of Trolox and ascorbic acid have a beneficial effect on in vitro maturation of pig oocytes

: The poor in vitro development of pig oocytes and embryos has been blamed on oxidative stress. We sought to find out if combinations Trolox (T), a synthetic cell-permeable derivative vitamin E, ascorbic acid (AA) could improve the maturation rates cultured oocytes. Pig underwent for 44–45 h medium M 199 supplemented with 0 μM T + AA, 100 250 300 750 AA or AA. These were chosen based previous research conducted our laboratory available literature. After maturation, several parameters assessed: cumulus oophorus expansion, oocyte viability (based presence metabolic activity versus membrane damage), extrusion first polar body, mitochondrial potential (MMP), pronucleus formation, embryo after fertilization. All antioxidant significantly improved expansion formation body. best was characteristic second. Antioxidant media increased percentages viable but not significantly. MMP modified by addition combinations. also found that low concentration (100 µM) mixed high (750 led higher both female male pronuclei enhanced morula stage. Therefore, we recommend this combination
